چکیده

Novel synthetic mimics of antimicrobial peptides have been developed to exhibit structural properties and antimicrobial activity similar to those of natural antimicrobial peptides (AMPs) of the innate immune system. These molecules have a number of potential advantages over conventional antibiotics, including reduced bacterial resistance, cost-effective preparation, and customizable designs. In this study, we investigate a family of nylon-3 polymer-based antimicrobials. By combining vesicle dye leakage, bacterial permeation, and bactericidal assays with small-angle X-ray scattering (SAXS), we find that these polymers are capable of two interdependent mechanisms of action: permeation of bacterial membranes and binding to intracellular targets such as DNA, with the latter necessarily dependent on the former. We systemically examine polymer-induced membrane deformation modes across a range of lipid compositions that mimic both bacteria and mammalian cell membranes. The results show that the polymers' ability to generate negative Gaussian curvature (NGC), a topological requirement for membrane permeation and cellular entry, in model Escherichia coli membranes correlates with their ability to permeate membranes without complete membrane disruption and kill E. coli cells. Our findings suggest that these polymers operate with a concentration-dependent mechanism of action: at low concentrations permeation and DNA binding occur without membrane disruption, while at high concentrations complete disruption of the membrane occurs. Epidermal mucus, a major determinant in fish health: a review

Fish epidermal mucus contains innate immune components, secreted by globlet cells that provide the primary defence against different pathogenic microbes and act as a barrier between fish and its immediate niche. The major function of mucus includes entrapment and sloughing of microbes. The mucus also contains many factors such as antimicrobial peptides (AMPs), lysozymes, lectins, proteases, etc...

A critical evaluation of random copolymer mimesis of homogeneous antimicrobial peptides.

Polymeric synthetic mimics of antimicrobial peptides (SMAMPs) have recently demonstrated similar antimicrobial activity as natural antimicrobial peptides (AMPs) from innate immunity. This is surprising, since polymeric SMAMPs are heterogeneous in terms of chemical structure (random sequence) and conformation (random coil), in contrast to defined amino acid sequence and intrinsic secondary struc...

Functional synergy between antimicrobial peptoids and peptides against Gram-negative bacteria.

Antimicrobial peptides (AMPs) are integral components of innate immunity and are typically found in combinations in which they can synergize for broader-spectrum or more potent activity. Previously, we reported peptoid mimics of AMPs with potent and selective antimicrobial activity. Using checkerboard assays, we demonstrate that peptoids and AMPs can interact synergistically, with fractional in...

Cationic Peptide Interactions with Biological Macromolecules

Cationic amphiphilic peptides (CAPs) are widely studied as effectors that are activated by microbial pathogens in immune signaling pathways of invertebrates and vertebrates. These peptides are non-specific effectors that can kill bacteria, fungi, viruses and protozoan parasites [1, 2]. They are a universal feature in all forms of life and are often found in all the major barriers such as the sk...

Antimicrobial Peptides of Innate Immune System as a Suitable Compound for Cancer Treatment and Reduction of its Related Infectious Disease

Application of chemotherapy in cancerous children leads to reduction of immune system efficiency. Therefore, these children are prone to various infectious diseases. The excessive use of antibiotics can bring about antibiotic resistant strains. Hence, it is essential to investigate new therapies for this problem. On the other hand, the emergence of resistance against multiple drugs is a major p...
